After the success of the first Ghostbusters film a second was all but guaranteed. Again produced and directed by Ivan Reitman and follows a Ghostbusters after a large number of bills from their battle with Gozer has bankrupted their business and forced them all to take on side jobs to keep themselves afloat.
The Ghostbusters team is in disarray after being held financially responsible for all the damage that occurred in their fight against Gozer and the Stay Puft Marshmallow Man. The city has also restrained the team from battling any paranormal ghosts or access to any of their equipment. Ray Stantz, who is running an occult bookstore, is still teaming up with Winston Zeddemore going around as the Ghostbusters for kid's birthday parties. Egon Spengler has been researching human emotional response. Peter Venkman is hosting a local television show called "World of the Psychic" where he pretends to read minds and garners little attention. Dana Barrett is now working at the Manhattan Museum of Art where she restores paintings raises her baby, Oscar.
One day Dana's stroller mysteriously gets pushed and controlled down a busy New York street putting Oscar in danger. This scares Dana enough to ask for help from the Ghostbusters even if that means reuniting with her ex-boyfriend, Venkman. At Dana's job though her boss, Dr. Janosz Poha has been possessed and comanded by Vigo the Carpathian who exists in a painting recently acquired by the museum. Vigo tells Poha that he needs Dana's child so that he can take over the child's body and get a physical form.
The Ghostbusters begin looking into Oscar's possibly paranormal influence and find that the influence has its origins from underneath the city. The four of them start at the same street in which Oscar stopped when he went through the city in his stroller earlier. Slowly lowered down Ray discovers one of the abandoned subway line is completely filled with pink slime flowing like a river. When trying to collect a sample Ray is seemingly attacked by the slime and turns off the power to most of the city.
The team is arrested and go to court where Louis Tully does defend them but to no avail and they are found guilty. During the trial their judge has several episodes of anger and the slime sample from the subway has a reaction to his emotions. The slime releases two ghosts who the judge had once sentenced to death. As the ghosts wreck the courtroom the judge lets the Ghostbusters trap the ghosts in exchange to the charges being dismissed and allow them to do their old jobs rescuing the city from ghosts.
With the Ghostbusters back in business the slime doesn't rest either, in Dana's apartment the slime attempts to kidnap Oscar. With nowhere else to go Dana goes to Peter for safety. The Ghostbusters begin finding slime all over the city which has been growing stronger with the negative emotions of all the citizens who live there. They also investigate the Vigo painting at Dana's work and figure that its ancient tyrant who once ruled. As Peter and Dana are going out on a date, the other Ghostbusters go further into the sewer system to investigate the slime more. Despite falling in and having the slime induce anger fits they discover all the slime is all flowing towards one place, the museum.
The Ghostbusters rush to the mayor's office to warn the officials only to be thrown into a mental institution. One evening Dana finds Oscar wandering out onto the ledge of the apartment and just before she can reach him her baby is kidnapped by a ghost. Dana runs to the museum to find Oscar there but is quickly sealed in my the growing slime that has now enveloped the museum. Dana's encounter with a ghost isn't an isolated incident though, soon all of New York is having the slime take over and ghosts going crazy. After the pure energy from the slime blocks the sun and the city is in complete panic mode the mayor releases the Ghostbusters.
At the museum the Ghostbuster's can't break through the wall of slime and the general negative energy from the citizens of New York is keeping the slime strong. They need something that can inspire all of the citizens and make them happy again. The Ghostbusters take a newly developed slime, created by Egon, and coat the inside of the Statue of Liberty. While playing one of the songs that the slime reacts to positively they are able to control the Statue of Liberty and with the new positive attitudes of the people, break the ceiling of the museum.
Using the positive slime Janosz is freed from his possession but Vigo seems able to fight off its effects. Vigo beats back the Ghostbusters and is able to move his spirit into Oscar's body. But as New Years approaches the citizens outside begin singing "Auld Lang Syne" and begins to weaken Vigo giving the Ghostbusters enough time to defeat him back into his painting. The Ghostbusters have saved the day and Vigo is destroyed, his painting replaced with baby Oscar and his four heroes.
